Introduction
Cas12b nuclease (also known as C2c1) is an RNA-mediated endonuclease, target double stranded DNA can be cleaved specifically in the presence of target DNA PAM. Cas12b protein is smaller than Cas9 and Cas12a. Aac Cas12b is derived from Alicyclobacillus acidoterrestris. Aap Cas12b Derived from Alicyclobacillus Acidophilus acidophilic. Aap Cas12b was more suitable for the one-tube system combined with LAMP. Cas12b has the best cutting reaction temperature of 48 ℃. Similar with Cas12a from the same family, Cas12b recognizes PAM sequences of 5'-TTN and cleaves DNA to produce sticky ends. However Cas12b is double RNA Guidance, dependent on crRNA and tracrRNA (or sgRNA formed by linking). In addition, Cas12b can be applied to more than just targets DsDNA cutting. Cas12b can also be used for rapid detection of target nucleic acid.
